Paypal Pro isn’t available for my country (Singapore). However we’re looking at Paypal Express & Subscriptions, does is it possible to have paypal express and subscriptions when paypal pro isn’t available to me?

Additionally, do I require S2Member Pro for this?

It appears that you’ve already purchased s2Member® Pro, so I would highly recommend that you take advantage of s2Member’s Pro Form integrations. You can do this with or without a PayPal® Pro account.
