I only need to protect the page single.php in wordpress with s2member.

Where should I do this or how do I do this? Thanks in advance

You might want to try URI access restrictions:
Dashboard -› s2Member® -› Restriction Options -› URI Access Restrictions

You might also be interested in post access restrictions:
Dashboard -› s2Member® -› Restriction Options -› Post Access Restrictions

And also:
Dashboard -› s2Member® -› Restriction Options -› Specific Post/Page Access Restrictions
